The increases in Scr and RI of renal blood vessels induced by diattizoate were suppressed significantly and dose-dependently by pretreatment with KB-R7943. Histopathological and TUNEL results showed that the
contrast medium-induced severe renal tubular necrosis and apoptosis were significantly and dosedependently attenuated by KB-R7943. KB-R7943 significantly suppressed the increment of renal ET-1 content and MDA and CAT level induced by contrast medium administration.
Conclusion: Activation of the reverse mode of NCX, followed by ET-1 overproduction and increased oxi[ dative stress, seems to play an important Fosbretabulin role in the pathogenesis of CI-AKI. The inhibitor of the reverse mode of NCX, KB-R7943, has renoprotective effects
on CI-AKI.”
“Background: It is known that recognition of bacterial lipopolysaccharide (LPS) and various endogenous ligands by Toll-like receptor 4 (TLR4) induces inflammatory reactions. However, the role of TLR4 activation in mesangial inflammation remains to be elucidated. Retinoic acid-inducible gene-I (RIG-I) and melanoma differentiation-associated gene 5 (MDA5) are putative RNA helicases and are involved in immune and inflammatory reactions. The purpose of the present study was to investigate the implication of RIG-I and MDA5 in TLR4 signaling in mesangial cells.
Methods: Normal human mesangial cells in culture were treated with LPS. Expression of RIG-I, MDA5, interferon-13 (IFN-13), CXCL10 and CXCL8 was examined using real-time RT-PCR, Western blotting and ELISA. The cells were also subjected to RNA FDA-approved Drug Library nmr interference against TLR4, IFN-(3, RIG-I or MDA5.
Results: LPS induced the expression of IFN-(3, RIG-I, MDA5, CXCL8 and CXCL10 in human mesangial cells. RNA interference against either TLR4 or IFN-13 inhibited LPS-induced RIG-I and
MDA5 expression. Knockdown of TLR4, RIG-I or MDA5 resulted in decreased induction of CXCL10, while only TLR4 knockdown inhibited CXCL8 induction.
Conclusions: TLR4 signaling induces the expression of RIG-I and MDA5 in mesangial cells. RIG-I and MDA5 may be involved in inflammatory reactions by regulating CXCL10 expression in the downstream of TLR4 signaling in human mesangial cells.”
“A check details new microfocused Brillouin light scattering (BLS) apparatus has been setup using a microscope objective with large numerical aperture and achieving a submicrometric lateral resolution, as inferred by reflectivity measurements taken on a set of calibration arrays consisting of Al stripes with different spacings. The micro-BLS apparatus has been then used to investigate the magnetic normal modes of elliptical Permalloy nanorings. The superiority of micro-BLS over conventional BLS measurements in the detection of stationary modes, especially those localized close to the edges of the nanoring where the internal field is highly nonhomogeneous, is demonstrated.
